BioFX® Chemiluminescent HRP Substrates

Five chemiluminescent substrates with luminol-based formulations are available for HRP detection. These products offer ultra-high or routine detection with the potential for from picogram to femtogram sensitivity. These products are stable for a minimum of 18 months at 2°C to 8°C and are easy-to-use, two-component formulations.

Shelf Life

These chemiluminescent substrates are stable for a minimum of 18 months from the date of manufacture

Mixtures of the two components (Reagents A and B) are stable for several hours at room temperature when protected from light

Product Use 

Mix well and protect from light

Mix 1 mL of Reagent A with 2 mL Reagent B

Larger and smaller volumes of the substrate can be mixed at this same 1:2 ratio

Mix well and protect from light

Mix 1 mL of Reagent A with 1 mL Reagent B

Larger and smaller volumes of the substrate can be mixed at this same 1:1 ratio

NOTE

These chemiluminescent substrates are very sensitive and great care must be taken to optimize the individual assay components (e.g., antibodies, solid phase, etc.) to minimize background reactivity associated with nonspecific immunochemical reactions.

Detection Limit Comparison: BioFX HRP Microwell Substrate vs. Competitor



Rabbit anti-HRP was diluted and coated to the plate. Plate was sealed and incubated at 37°C for 1 hour then washed 3X with PBST. A diluted HRP standard curve was added to the plate. Standard curve ranged from 1000 pg/well to 15.625 pg/well. Plate was incubated for 1 hour at 37°C then washed 3X with PBST. Various substrates were added to appropriate wells. The plate was then incubated for 10 min. at R.T. prior to reading.

BioFX HRP Membrane Substrate



Mouse IgG was run on a 4%-20% reducing gel. Bands were transferred to a nitrocellulose membrane, blocked for 1 hour at 4°C, and incubated with HRP-conjugated rabbit anti-mouse IgG for 1 hour. After washing in PBST, BioFX chemiluminescent substrate (CHMM) was added and the blot visualized using a CCD camera. Note the low background and good contrast in the resulting image.
