BioFX® DAB HRP Membrane Substrate

BioFX® DAB HRP Membrane Substrate increases stability and greatly improves sensitivity for rapid and simple detection of peroxidase in membrane applications.

BioFX DAB Membrane Substrate (Product Code DABM)
Product Description
  • Supplied as a two-component liquid substrate
  • DABC - Membrane Concentrate (Reagent A)
  • DABD - Membrane Reaction Buffer (Reagent B)
  • Reaction with peroxidase results in a light golden brown to dark brown precipitate
How it Works
  • DAB (3,3'-diaminobenzidine) is oxidized in the presence of peroxidase and hydrogen peroxide resulting in the deposition of a brown, alcohol-insoluble precipitate at the site of enzymatic activity
Shelf Life
  • Stable for a minimum of 3 years from the date of manufacture when stored at 2°C - 8°C in the original containers
  • After combination of the two reagents, the resulting 1X solution is stable for approximately 2 to 4 hours at room temperature (25°C)
  • The 1X solution may darken during this 2 - 4 hour time frame; this will not affect product performance
Storage
  • Store at 2°C - 8°C in the original container away from light and heat with lid tightly closed

Western Blot using DAB HRP Membrane Substrate – Comparisons were performed on western blots detecting serial dilutions of HSP 70 (10 to 0.04 μg) with anti-HSP-70 at a 1:2K dilution. Blots were washed 3X. A biotin-labeled Goat Anti-Mouse IgG was used as the secondary at 1:1K diltuion, Streptavidin-HRP was used at 1:1.5K with detection by BioFX DABM.
